When trailering long distances (e.g., to Toledo Bend, Lake Fork, etc.), which is more fuel efficient?

A. with boat cover on

B. with boat cover off

I'm not sure about fuel efficiency but if the weather is nasty I will put the cover on. If the weather is nice I roll with the cover off.

If you have a custom cover that is tied down really tight and it doesnt sag while trailering it then you will get about the same fuel efficiency with or without the cover on. If your cover saga like my old canvas one, then you will get worse fuel efficiency bc the cover acts like a giant wind sock.
